Helicase Unzips, Primase Preps, Pol-Three Synthesizes, Pol-One Erases, Ligase Glues
Picture This (Mental Hook): Think of a factory assembly line: Helicase the brawny worker unzips the heavy DNA zipper. Primase leaves RNA sticky notes. DNA Polymerase III speeds along building new DNA tracks. DNA Polymerase I rips out the sticky notes and fills the gaps. Finally, Ligase glues the Okazaki seams shut with molecular tape!

[Primer] [Primer] [Primer]
* [SSB Proteins] prevent re-annealing and hairpin formation
* [Topoisomerase / Gyrase] relieves torsional supercoiling ahead of the fork
* [Pol I] has 5' โโโบ 3' EXONUCLEASE activity to degrade RNA primers!๐ The Decoded Sequence:
Helicase
Unwinds
DnaB hexamer in E. coli tracking along lagging strand
Gyrase
Untwists
Type II topoisomerase cutting both strands to relieve positive supercoils
Pol III
Main Replicator
High processivity holoenzyme with beta-sliding clamp
Pol I
Proofreader & Primer Remover
Unique 5'->3' exonuclease Nick Translation activity
Ligase
Phosphodiester Seal
Catalyzes phosphodiester bond formation between 3'-OH and 5'-PO4
Examiner Trap / Never Forget: DNA Polymerase III has 3'->5' exonuclease proofreading activity, but LACKS 5'->3' exonuclease activity. ONLY DNA Polymerase I has 5'->3' exonuclease activity to degrade RNA primers!
