Pollen Viability Assessment & In Vitro Pollen Tube Germination Kinetics in Brewbaker-Kwack Medium
Lab Duration: 3 Hours • Catharanthus roseus / Hibiscus • TTC & Hanging Drop
๐ฏ Aim: To evaluate pollen viability using the tetrazolium (TTC) enzymatic reduction assay and acetocarmine staining, germinate pollen grains of *Catharanthus roseus* (*Vinca*) in Brewbaker and Kwack liquid culture medium using hanging drop culture, determine germination percentages, and measure tip-focused pollen tube elongation kinetics ($mutext{m/hr}$) over time.
๐ฌ Principle & Tip-Growth Mechanics
In angiosperms, successful sexual reproduction depends on pollen viability (the metabolic capacity of the male gametophyte to germinate and deliver non-motile sperm cells to the female ovule):
- Tetrazolium (TTC) Viability Assay: Living, metabolically active pollen contains functional mitochondrial dehydrogenase enzymes (succinate dehydrogenase). These dehydrogenases reduce colorless, water-soluble 2,3,5-triphenyltetrazolium chloride (TTC) into insoluble, bright carmine red formazan crystals. Non-viable or dead pollen lacks enzyme activity and remains completely colorless.
In Vitro Germination & Tip-Focused Growth:
- When placed in appropriate osmotic nutrient media, hydrated pollen grains germinate by pushing an intine-derived tubular protrusion out through an aperture (germ pore) in the sculptured sporopollenin exine wall.
- Pollen tube elongation exhibits one of the fastest rates of tip-focused polarized growth known in plant biology (up to $10 – 20,mutext{m/min}$ in vivo). Secretory vesicles derived from Golgi bodies accumulate in the apical dome (“clear zone”), fusing with the tip membrane to deliver pectin and cellulose precursors for cell wall synthesis.
- The classical Brewbaker and Kwack (BK) medium provides essential ionic and osmotic cofactors:
– Sucrose ($10%,text{w/v}$): Provides precise osmotic support (preventing osmotic bursting of pollen grains) and an energy carbon source.
– Boric Acid ($100,text{mg/L},text{H}_3text{BO}_3$): Borate complexes with sucrose to form sugar-borate complexes that promote pectin synthesis and crosslink rhamnogalacturonan-II in the pollen tube wall.
– Calcium Nitrate ($300,text{mg/L},text{Ca(NO}_3)_2$): Maintains a steep intracellular tip-focused calcium gradient ($[text{Ca}^{2+}]_i > 10,mutext{M}$ at the apex vs $< 200,text{nM}$ in the shank), directing exocytosis and vesicle fusion. - Magnesium Sulfate ($200,text{mg/L}$) & Potassium Nitrate ($100,text{mg/L}$): Support membrane transport and turgor maintenance.

๐ Step-by-Step Germination & Kinetics Protocol
- Brewbaker & Kwack (BK) Medium Formulation: Prepare $100,text{mL}$ of standard BK germination medium: dissolve $10.0,text{g}$ sucrose, $10,text{mg},text{H}_3text{BO}_3$, $30,text{mg},text{Ca(NO}_3)_2cdot 4text{H}_2text{O}$, $20,text{mg},text{MgSO}_4cdot 7text{H}_2text{O}$, and $10,text{mg},text{KNO}_3$ in distilled water. Adjust pH to $6.8 – 7.0$ and filter.
- TTC Viability Staining: Dust fresh mature anthers of *Catharanthus roseus* onto a glass slide containing a drop of $1%,text{TTC}$ in $0.2,text{M}$ phosphate buffer ($text{pH } 7.2$). Apply coverslip and incubate in the dark at $35^circtext{C}$ for 30 minutes. Examine under $100times$: count viable grains (stained bright pink/carmine red) and non-viable grains (colorless). Calculate:
$$% text{ Viability} = left( frac{N_{text{red}}}{N_{text{total}}} right) times 100$$ - Hanging Drop Germination Assembly: Place a drop of BK medium ($20,mutext{L}$) on a clean coverslip. Gently dust pollen grains from dehisced *Catharanthus* anthers into the droplet using a camel-hair brush. Invert the coverslip over a cavity slide rimmed with paraffin oil or moist filter paper (forming a humid chamber to prevent evaporation).
- Incubation & Scoring Criteria: Incubate at $25^circtext{C}$ in the dark. Examine under brightfield ($100times$) at 30, 60, 90, and 120 minutes. A pollen grain is scored as germinated when the emerging pollen tube length exceeds the diameter of the pollen grain. Count at least 150 grains across 5 fields. Calculate:
$$% text{ Germination} = left( frac{N_{text{germinated}}}{N_{text{total}}} right) times 100$$ - Tube Elongation Rate Measurement: Using an ocular micrometer calibrated with a stage micrometer, measure the absolute length ($L$ in $mutext{m}$) of 15 individual pollen tubes at 15-minute intervals. Plot tube length versus incubation time. Calculate the linear growth velocity ($V = Delta L / Delta t$) during the exponential growth phase ($sim 150 – 300,mutext{m/hr}$).
