Separation of Foliar Chloroplast Pigments via Paper & Thin-Layer Chromatography (TLC): Rf Determination

Lab Duration: 3 Hours • Spinacia oleracea (Spinach) • Petroleum Ether : Acetone (9:1)
๐ŸŽฏ Aim: To extract crude photosynthetic pigment complexes from fresh spinach (*Spinacia oleracea*) leaves, resolve individual pigments (Chlorophyll a, Chlorophyll b, Xanthophylls, Carotenes, and Pheophytin) using ascending paper and thin-layer chromatography, and determine Retention Factor ($R_f$) values based on differential molecular polarities.

๐Ÿ”ฌ Principle & Physicochemical Partitioning

Photosynthetic thylakoid membranes contain a mixture of lipid-soluble pigments responsible for light harvesting and photoprotection:

  • Chlorophyll a ($text{C}_{55}text{H}_{72}text{O}_5text{N}_4text{Mg}$): Primary reaction center pigment; blue-green in color; possesses a methyl group ($-text{CH}_3$) at carbon-7 of the porphyrin ring.
  • Chlorophyll b ($text{C}_{55}text{H}_{70}text{O}_6text{N}_4text{Mg}$): Accessory antenna pigment; yellow-green in color; possesses an electronegative formyl group ($-text{CHO}$) at carbon-7, rendering it more polar than chlorophyll a.
  • Carotenoids:
    – Carotenes ($beta$-Carotene, $text{C}_{40}text{H}_{56}$): Orange-yellow pure hydrocarbons containing conjugated double bonds without oxygen functional groups; highly non-polar.
    – Xanthophylls (Lutein/Violaxanthin, $text{C}_{40}text{H}_{56}text{O}_2$): Yellow oxygenated carotenoids containing hydroxyl groups ($-text{OH}$); intermediate polarity.

  • Pheophytin: A magnesium-free chlorophyll breakdown product formed by weak acid displacement of central $text{Mg}^{2+}$, appearing grey-olive.

In ascending partition chromatography, the stationary phase consists of highly polar cellulose fibers (adsorbed water layer on paper) or silica gel ($text{SiO}_2$). The mobile phase is a relatively non-polar solvent mixture: petroleum ether : acetone ($9:1,text{v/v}$).

As the mobile phase ascends by capillary action, pigments partition continuously between the stationary and mobile phases:

  • Non-polar pigments (Carotenes) have high solubility in petroleum ether and minimal affinity for polar cellulose, traveling near the solvent front ($R_f approx 0.95$).
  • Polar pigments (Chlorophyll b) form strong hydrogen bonds with cellulose hydroxyls and partition poorly into the non-polar solvent, migrating slowly near the origin line ($R_f approx 0.25$).

The relative migration distance is expressed as the Retention Factor ($R_f$):
$$R_f = frac{text{Distance migrated by the pigment band from the origin (cm)}}{text{Distance migrated by the solvent front from the origin (cm)}}$$

๐Ÿ“‹ Step-by-Step Chromatography Protocol

  1. Pigment Extraction: Grind $5.0,text{g}$ of fresh spinach leaves in a chilled mortar with $10,text{mL}$ of $80%$ acetone, a pinch of acid-washed sand (to rupture cell walls), and $0.1,text{g}$ of $text{CaCO}_3$ (to neutralize plant acids and prevent chlorophyll pheophytinization). Filter the extract through Whatman #1 filter paper into a brown glass vial to protect against photodegradation.
  2. Paper Preparation: Cut a $20 times 3,text{cm}$ strip of Whatman #1 chromatography paper. Draw a faint pencil origin line $2.0,text{cm}$ from the bottom edge. Cut the bottom edge into a tapered “V” shape below the origin line to ensure uniform solvent ascent.
  3. Sample Spotting: Using a fine glass capillary tube, apply a tiny drop of dark green pigment extract onto the center of the origin line. Allow it to air-dry completely. Repeat spotting $15 – 20$ times directly on the same spot until a dark, concentrated green dot ($< 3,text{mm}$ diameter) is formed.
  4. Chamber Development: Pour $15,text{mL}$ of running solvent (petroleum ether $60-80^circtext{C}$ : acetone in $9:1$ ratio) into a tall glass chromatography jar. Swirl and allow the chamber atmosphere to saturate for 15 minutes. Suspend the paper strip vertically with a cork hook so that the tapered tip dips $5,text{mm}$ into the solvent, ensuring the origin spot remains strictly above the liquid level.
  5. Elution & Marking: Allow solvent to ascend in the dark for 25 to 35 minutes until the solvent front is $sim 1,text{cm}$ below the top edge. Remove the strip immediately, quickly mark the solvent front with a pencil before it evaporates, and circle the center of each separated colored band.
  6. Rf Measurements: Measure the distance from the origin line to the solvent front and to the center of each pigment band using a vernier ruler. Calculate $R_f$ values for all 4 distinct bands:
    – $beta$-Carotene (bright orange, top, $R_f approx 0.95$)
    – Xanthophyll (yellow, second, $R_f approx 0.65$)
    – Chlorophyll a (blue-green, third, $R_f approx 0.45$)
    – Chlorophyll b (olive-green, bottom, $R_f approx 0.25$)
