Quantitative Protein Estimation by Lowry and Bradford Spectrophotometric Assays
๐ฏ Aim: To construct standard calibration curves using Bovine Serum Albumin (BSA) by the Lowry (Folin-Ciocalteu) and Bradford (Coomassie Brilliant Blue G-250) assays, compare their linear detection limits, and quantitatively determine the protein concentration of an unknown biological extract.
๐ฌ Theoretical Principle & Optical Mechanics
Accurate quantification of protein concentration is fundamental to all enzyme kinetics, molecular biology, and biochemical purification protocols:
- Lowry Method (Folin-Ciocalteu Assay): Combines the Biuret reaction with the reduction of the Folin-Ciocalteu phenol reagent (phosphomolybdate and phosphotungstate). In alkaline copper solution, peptide bonds chelate $text{Cu}^{2+}$ to form $text{Cu}^+$. Monovalent copper, along with tyrosine, tryptophan, and cysteine side chains, reduces the yellow phosphomolybdic-phosphotungstic acid complex to an intense molybdenum/tungsten blue chromophore measured at $lambda = 660,text{nm}$. Sensitivity: $5–100,mutext{g/mL}$.
- Bradford Method (Dye-Binding Assay): Employs Coomassie Brilliant Blue G-250. In acidic medium, the dye exists in a doubly protonated red/brown cationic state ($lambda_{max} = 465,text{nm}$). Upon binding to basic (primarily arginine, lysine, histidine) and aromatic amino acid residues of proteins, the dye is converted to an unprotonated blue anionic sulfonate form ($lambda_{max} = 595,text{nm}$). The absorbance at $595,text{nm}$ is directly proportional to protein concentration via the Beer-Lambert law ($A = varepsilon cdot c cdot l$). Sensitivity: $1–50,mutext{g/mL}$.

๐ BSA Bradford Assay Pipetting Protocol
| Tube No. | Standard BSA (100 μg/mL) | dH2O (μL) | BSA Protein (μg) | Bradford Reagent (mL) | Incubation (min) | Mean A595 |
|---|---|---|---|---|---|---|
| Blank (B) | 0 μL | 100 μL | 0.0 μg | 5.0 mL | 5 min at RT | 0.000 |
| S1 | 20 μL | 80 μL | 2.0 μg | 5.0 mL | 5 min at RT | 0.082 |
| S2 | 40 μL | 60 μL | 4.0 μg | 5.0 mL | 5 min at RT | 0.165 |
| S3 | 60 μL | 40 μL | 6.0 μg | 5.0 mL | 5 min at RT | 0.248 |
| S4 | 80 μL | 20 μL | 8.0 μg | 5.0 mL | 5 min at RT | 0.330 |
| S5 | 100 μL | 0 μL | 10.0 μg | 5.0 mL | 5 min at RT | 0.412 |
| Unknown (U) | 50 μL | 50 μL | — | 5.0 mL | 5 min at RT | 0.215 |
Calculation of Unknown Sample:
Using the linear regression equation $y = mx + c$:
$$text{Protein Conc. } (mutext{g/mL}) = frac{(A_{595} – c)}{m} times frac{1}{text{Volume of sample in mL}} times text{Dilution Factor}$$
$$text{For } A_{595} = 0.215: quad text{BSA Equivalent} = frac{0.215}{0.0412} = 5.22,mutext{g} implies frac{5.22,mutext{g}}{0.05,text{mL}} = 104.4,mutext{g/mL}$$

๐ฌ Viva-Voce Questions & Model Answers
Q1: What are the primary interfering substances in the Lowry and Bradford assays?
The Lowry assay is heavily compromised by reducing agents (DTT, $beta$-mercaptoethanol), chelating agents (EDTA), and detergent buffers (Triton X-100), which prematurely reduce the Folin reagent. Conversely, the Bradford assay is unaffected by reducing agents but is severely distorted by high concentrations of detergents (SDS, Triton X-100) which bind the Coomassie dye and precipitate it.
Q2: Why must the Bradford absorbance be read within 10–60 minutes?
The Coomassie dye-protein complex remains stable in solution for approximately 1 hour. After prolonged incubation, the dye-protein complex aggregates and precipitates out of solution, leading to loss of absorbance and non-linear baseline drift.
Q3: Why is Bovine Serum Albumin (BSA) chosen as the universal calibration standard?
BSA is exceptionally stable, highly soluble, commercially available in ultrapure crystalline form at low cost, and contains an average distribution of basic, aromatic, and neutral amino acids that closely resembles representative mammalian proteins.
